What to know before you buy. We would rather tell you this than have you find out later: AOD-9604 was tested properly in placebo-controlled human obesity trials, and it did not beat placebo on weight loss. That is unusual in this category, where most compounds have never been tested at all — and it cuts both ways, because it also means the safety data is better than most. It was well tolerated and did not disturb blood sugar or IGF-1. If you want it for tolerability or for the newer joint and cartilage research, that is a defensible reason. If you want it for fat loss, the trial record does not support that.

Evidence status — read this first

AOD-9604 is one of the few compounds in this category to have completed proper placebo-controlled human obesity trials. It did not produce statistically significant weight loss versus placebo on the primary endpoint, and the obesity programme did not advance to approval. Marketing that presents AOD-9604 as a proven fat-loss compound is not consistent with its own trial record. It was not on the July 2026 PCAC agenda.

What the Trials Showed

The development programme took AOD-9604 through phase II in obesity. The trials achieved something most compounds here never attempt: a real placebo-controlled test of the central claim. The headline result was that the compound did not separate meaningfully from placebo on weight reduction.

The safety findings were genuinely favourable. AOD-9604 did not produce the IGF-1 elevation or glucose effects associated with full-length growth hormone, which is consistent with the design rationale — the fragment does appear to avoid those effects. It simply also appears to avoid producing the intended one at a clinically meaningful level.

Why it remains on the market

Interest persisted partly on the strength of preclinical work and partly through subsequent attention to other possible applications, including cartilage and joint research. Those lines are early and are not the basis on which the compound is usually sold. Its favourable tolerability profile also keeps it in circulation.

The bottom line

A well-designed idea that was properly tested and did not deliver on its primary endpoint. Reliable tolerability, no demonstrated fat-loss efficacy versus placebo. Of everything in this range, AOD-9604 is the compound where the honest evidence summary differs most sharply from the typical sales pitch.
